NAD+ (nicotinamide adenine dinucleotide) is the universal redox cofactor whose tissue-pool depletion is repeatedly observed in ageing-biology literature; sirtuin-family deacetylases (SIRT1-7), PARPs, and CD38 all draw from it, and NAD+ research has been a defining theme of the last decade of senescence work.
Glutathioneis the cell’s principal water-phase antioxidant — a tripeptide (γ-Glu-Cys-Gly) maintained at millimolar concentrations in the cytosol. Its thiol-disulfide chemistry sits at the heart of every published redox-balance assay. Note the practical caveat: glutathione is oxidation-sensitive, so reconstituted research preparations have shorter stability windows than typical peptide cofactors.
MOTS-cis the mitochondrial-derived peptide encoded within the 12S rRNA region of the mtDNA. Discovered by the Cohen lab and characterised primarily in rodent metabolic and insulin-sensitivity models, it’s the most prominent member of the mitochondrial-derived-peptide (MDP) family in current research. Plasma half-lives across this shelf are short (minutes for both NAD+ and glutathione; not well-characterised for MOTS-c), and protocols vary substantially by route.
