CJC-1295 without DAC, also referenced in the literature as Modified GRF (1-29), is a tetrasubstituted analogue of the first 29 residues of human growth hormone-releasing hormone (hGRF 1-29). The four backbone substitutions (D-Ala2, Gln8, Ala15, Leu27) were originally introduced to limit dipeptidyl peptidase IV cleavage and trypsin-like degradation observed for native GRF(1-29). Unlike the DAC-conjugated variant, the unconjugated peptide is not covalently linked to a maleimido-lysine carrier and is characterised in preclinical work as a short-acting GHRH-receptor agonist. Published in vitro and animal-model studies have examined its interaction with the pituitary GHRH receptor, the pulsatile somatotropic axis, and its analytical fingerprint in doping-control matrices. Published protocols
Alba et al. 2006 (Am J Physiol Endocrinol Metab) used once-daily SC administration in GHRH-knockout mouse models. The Teichman et al. 2006 (JCEM) human trial examined SC doses of 30–60 µg/kg as single administrations; the short-acting (no DAC) analogue was not the primary subject but served as reference context for half-life comparisons.

~30 minutes in vivo due to rapid DPP-IV cleavage; significantly shorter than the DAC-conjugated variant. Characterised in Jetté et al. 2005 (Endocrinology) and Bai & Chang 1995 (J Pharm Pharmacol).
